It runs in a north-south direction starting from Outwater Drive and crosses Charlotte Street, Corinthia Street, Fletcher Street, Washington Street, Prentice Street, Phelps Street, Grand Street, Green Street, Caledonia Street, West Caledonia Street and Ontario Street. This street is part of the Bicycle route "Erie Canalway Trail - 1 Buffalo to Fairgrounds" as well as the Hiking trail "Erie Canalway Trail (Amherst to Lockport)". Prospect Street also exists in City of Niagara Falls, Town of Newfane and Town of Royalton.
