It runs in an east-west direction and crosses Bruce Street, Cheffers Street, Waverley Street, Huntly Street, Argyle Street, Norfolk Street, Lennox Street, Sussex Street, Bowen Street, Hudson Street, Darling Street and York Street. It ends in a cul-de-sac and connects Moonee Ponds and Aberfeldie. Holmes Road also exists in Yarra Junction and Red Hill.
