It runs in an east-west direction starting from Harrison Avenue and crosses Winchester Avenue, Kings Drive, Lexington Avenue, Savannah Highway, Saxton Avenue, Somerset Avenue, Saint Johns Avenue and Calhoun Avenue and ends at Virginia Avenue. Third Street is also known as S-38-645. Third Street also exists in Laurens County and Newberry County.
