It runs in a northeast-southwest direction starting from Smith Street and crosses Wright Street, Childers Street, Clarke Street, Campaspe Street, Cardwell Street and Margaret Street and ends at Midland Highway. Jeffrey Street also exists in City of Greater Geelong, Shire of Macedon Ranges, City of Frankston and Shire of South Gippsland.
